Effectiveness of Punica granatum Linn Extract Compared to Ellagic Acid on Inhibition of Oral Squamous Cell Carcinoma through Apoptotic Expression Analysis

Abstract:

At present cancer therapy using radiology and chemotherapy still leave problems, which may cause death in normal cells. One of the relatively safe alternative treatments is the utilization of Punica Granatum Linn (PGL). PGL extract and Ellagic Acid (EA) contained in PGL extract are active substances that can increase apoptosis. The purpose of this study was to compare the effectiveness of PGL and EA in enhancing apoptosis. 32 male rats; aged 5 months and weighed 30-50 grams divided into 4 groups. Two treatment groups (P1: exposed to benzopirene and treated with EA; P2: exposed to benzopirene and treated with PGL extract). Two control groups (K0: not exposed and without treatment: K1: exposed to benzopirene but without treatment). Biopsy was performed on the buccal mucosa that had undergone squamous cell carcinoma. Apoptosis was conducted using the Tunel test. Examination of cells undergoing apoptosis was carried out under microscope observation with 400 X magnification. There was a difference in the number of cell expression undergoing apoptosis. The largest average expression of apoptosis occurred in benzopirene exposure by PGL treatment (0.367 ± 0.196). The administration of PGL extract in squamous cell carcinoma is suspected to inhibit more the growth of cancer cells compared to EA by increasing apoptosis.
